目的 探讨核酸疫苗在 HCV感染中的预防和治疗价值 .方法 将 HCV C区基因插入 p EF质粒 EF- 1α启动子下游 ,构建 p EF- HCV C重组质粒 .然后经 im及皮下注射免疫 BAL B/ c小鼠 ,免疫次数分别为 1,2 ,3,4次 ,EL ISA法检测免疫小鼠血清中抗 - HCV的水平 .结果 接种后各组均检测到抗 - HCV,但是抗体滴度较低 .在不同的免疫次数中 ,4次免疫组抗体水平最高 .结论 构建的 HCV DNA重组体可诱发 BAL B/ c小鼠产生体液免疫应答 ,加强免疫后可使血清中抗 - HCV明显增高 .
Objective To investigate the prophylactic and therapeutic value of nucleic acid vaccine in HCV infection.Methods The HCV C region gene was inserted into the downstream of EF-1α promoter of pEF plasmid to construct p EF-HCV C recombinant plasmid.After immunization with BAL and BALB / c mice were immunized for 1, 2, 3 and 4 times respectively.The level of anti-HCV in serum was detected by ELISA.Results Anti-HCV was detected in all groups after vaccination, but the antibody titers were lower .In the different immunization times, the antibody level of the four immunized groups was the highest.Conclusion The recombinant HCV DNA recombinant can induce the humoral immune response in BALB / c mice and the serum anti-HCV level can be significantly increased after the immunization.
