目的应用生物信息学方法分析结核分枝杆菌Rv2629基因编码蛋白的结构和功能。方法从NCBI GenBank中获得结核分枝杆菌H37Rv利福平敏感菌株Rv2629的基因序列,利用在线分析软件预测和分析其编码蛋白的理化性质和亲疏水性,跨膜结构域及信号肽,糖基化位点及磷酸化位点,二级和三级结构及B、T淋巴细胞抗原表位。结果Rv2629蛋白由374个氨基酸组成,具有亲水性,主要存在于细胞壁及细胞膜。Rv2629含有信号肽及跨膜结构域,6个糖基化位点和4个磷酸化氨基酸位点,二级结构中α螺旋约占45.72%,β折叠约占17.11%。该蛋白含有3个潜在的B细胞抗原表位,其优势抗原表位主要集中在67-70,91-95,229-232位氨基酸,4个CTL细胞表位分别位于279、283、98、346位氨基酸,11个T h细胞表位分别位于26、127、256、25、34、116、204、215、350、363、367位氨基酸。结论Rv2629蛋白存在于细胞壁及细胞膜,是一分泌蛋白,在结核病的潜伏期可上调表达。该蛋白含有B、T细胞抗原表位,可作为结核利福平耐药株的研究靶点。
Objective To analyze the structure and function of the protein encoded by Mycobacterium tuberculosis Rv2629 using bioinformatics method. Methods The gene sequence of mycobacterium tuberculosis H37Rv rifampicin-sensitive strain Rv2629 was obtained from NCBI GenBank. The physical and chemical properties, hydrophilicity and hydrophobicity, transmembrane domain and signal peptide, glycosylation position Point and phosphorylation sites, secondary and tertiary structures and B, T lymphocyte epitopes. Results The Rv2629 protein consisted of 374 amino acids, which was hydrophilic and mainly existed in cell wall and cell membrane. Rv2629 contains signal peptide and transmembrane domain, 6 glycosylation sites and 4 phosphorylated amino acid sites. The secondary structure contains about 45.72% alpha helices and 17.11% beta-sheets. The protein contains three potential B cell epitopes, the dominant epitopes are mainly concentrated in amino acids 67-70,91-95,229-232, four CTL cell epitopes located at 279,283,98,346 amino acids The 11 T h cell epitopes are located at amino acids 26, 127, 256, 25, 34, 116, 204, 215, 350, 363, 367, respectively. Conclusion Rv2629 protein is present in the cell wall and cell membrane and is a secreted protein that can be up-regulated in the incubation period of tuberculosis. The protein contains B, T cell epitopes, which can be used as research targets of tuberculosis rifampicin resistant strains.
