目的探讨非霍奇金淋巴瘤(Non-Hodgkin′s Lymphoma,NHL)中p53基因第5~8外显子突变情况。方法采用PCR-SSCP法对47例NHL进行p53基因第5~8外显子的点突变研究。结果p53基因第5~8外显子总突变率为48.94%(23/47)。其中第6外显子的突变率最高为17.02%,其次为第8外显子,突变率为12.87%;高度恶性组和低度恶性组突变率分别为50.00%(12/24)、34.78%(8/23)(P>0.05);T细胞和B细胞突变率分别为39.29%(11/28)、47.37%(9/19)(P>0.05)。结论NHL中存在较高p53基因突变率,提示其突变可能参与了NHL的发生发展。高度恶性组p53基因突变倾向于高发。
Objective To investigate the mutation of exon 5 ~ 8 of p53 gene in non-Hodgkin’s Lymphoma (NHL). Methods The mutation of exon 5 ~ 8 of p53 gene in 47 cases of NHL was studied by PCR-SSCP. Results The total mutation rate of p53 exons 5-8 was 48.94% (23/47). The mutation rate of exon 6 was 17.02%, followed by exon 8, the mutation rate was 12.87%. The mutation rates of high grade group and low grade group were 50.00% (12/24) and 34.78% (8/23) (P> 0.05). The mutation rates of T cells and B cells were 39.29% (11/28) and 47.37% (9/19) respectively (P> 0.05). Conclusion There is a high mutation rate of p53 gene in NHL, suggesting that the mutation may be involved in the development of NHL. P53 gene mutations in highly malignant group tend to be high.
