目的探讨大鼠脑缺血后不同时间轴突生长抑制因子Nogo-A在远隔部位小脑皮质的动态表达。方法 60只大鼠随机分为假手术组(n=30只)和大脑中动脉闭塞组(n=30只),每组分为1、3、5、7和10 d五个不同的亚组;采用改良的ZeaLonga法制备大鼠大脑中动脉闭塞模型;HE染色观察小脑皮质组织形态学改变;免疫组化染色检测小脑皮质Nogo-A蛋白的表达。结果 Nogo-A蛋白免疫反应性细胞广泛分布于小脑皮层少突胶质细胞(OLG)构成的髓鞘区域,主要位于OLG(胞体小、胞浆少和突起少)的细胞浆,并于脑缺血后第5 d、7 d和10 d较假手术组明显增高(P<0.05),第1 d和3 d无统计学差异(P>0.05)。结论大鼠大脑中动脉闭塞后远隔部位小脑皮层Nogo-A主要由OLG表达且呈动态变化。
Objective To investigate the dynamic expression of neurite outgrowth inhibitory factor Nogo-A in the distal part of cerebellar cortex at different time points after cerebral ischemia in rats. Methods Sixty rats were randomly divided into sham operation group (n = 30) and middle cerebral artery occlusion group (n = 30), and each group consisted of five different subgroups The middle cerebral artery occlusion model was made by the modified ZeaLonga method. Morphological changes of cerebellar cortex were observed by HE staining. The expression of Nogo-A protein in cerebellum was detected by immunohistochemistry. Results Nogo-A protein immunoreactive cells were widely distributed in the myelin sheath composed of oligodendrocytes (OLG) in the cerebellum, mainly located in the cytoplasm of OLG (small cytoplasm, less cytoplasm and less protrusions) On the 5th, 7th, and 10th days after blood injection, the blood levels were significantly increased (P <0.05), but no significant difference was found on the 1st and 3rd days (P> 0.05). Conclusion Nogo-A in the distal part of cerebellar cortex after OLA occlusion is mainly expressed by OLG and shows dynamic changes.
