采用室内培养实验方法 ,研究了不同浓度和不同时间胺草磷 (APM )对蚕豆根系细胞有丝分裂及根系蛋白质和POD同工酶的影响。结果表明 :(1)不同浓度APM处理后 ,可明显提高有丝分裂指数 ,但与APM浓度的增加无明显线性关系。(2)5μmol/L浓度处理的根系 ,其蛋白质谱带出现增加与缺失现象 ,与对照相比 ,增加了4条谱带 ,在组蛋白区域缺失1条谱带。(3)根系内POD同工酶的变化趋势与蛋白质变化基本吻合 ,1~4μmol/L处理未呈现明显变化 ,但在5μmol/L下与对照相比诱导出一条活性很强的POD3 谱带。综上所述 ,5μmol/LAPM浓度处理可以认为是蚕豆根尖中一些生化性状产生变化的临界浓度。
The effects of APM on the mitosis, root protein and POD isoenzyme of Vicia faba root cells were studied by laboratory culture method at different concentrations and different times. The results showed that: (1) The mitotic index of APM treated with different concentrations of APM could be obviously increased, but there was no obvious linear relationship with the increase of APM concentration. (2) In the roots treated with 5μmol / L, the bands of the protein increased and decreased, compared with the control, four bands were added and one band was missing in the histone region. (3) The trend of POD isozymes in root system was in good agreement with the change of protein. No obvious change was observed in 1 ~ 4μmol / L treatment, but POD3 band with strong activity was induced at 5μmol / L. In conclusion, the concentration of 5μmol / L APM can be considered as the critical concentration of some biochemical traits in Vicia faba root tips.
