从光棘球海胆黄中提取纯化多糖,通过水提、Sevag除蛋白及醇沉得到海胆黄粗多糖,经DEAE-52阴离子交换柱层析,氯化钠梯度洗脱得到一种单一多糖组分SEP-S,再经Sephacryl S-400柱层析纯化得到SEP-S精品多糖。高效凝胶液相色谱鉴定其纯度可达97.6%。体外活性结果表明,SEP-S具有显著刺激小鼠脾淋巴细胞增殖的作用且呈剂量依赖性;同时SEP-S也具有抗肿瘤作用,在250μg/m L浓度时,其对人肺癌细胞A549和小鼠肝癌细胞H22生长的体外抑制率分别为31.1%及30.5%。研究结果表明,SEP-S体外具有良好的免疫调节作用及抗肿瘤作用。
Purified polysaccharides were extracted from the sea urchin of Echinochloa crusgalli, and crude polysaccharide of sea urchin was obtained by water extraction, Sevag deproteinization and alcohol precipitation, and then a single polysaccharide fraction was obtained by gradient elution on a DEAE-52 anion exchange column and sodium chloride SEP-S, and then purified by Sephacryl S-400 column chromatography to obtain SEP-S high quality polysaccharide. High-performance gel chromatography to identify its purity up to 97.6%. In vitro activity results showed that SEP-S could significantly stimulate the proliferation of mouse splenic lymphocytes in a dose-dependent manner. Meanwhile, SEP-S also had anti-tumor effect. At the concentration of 250μg / mL, SEP-S could inhibit the proliferation of human lung cancer cells A549 and The inhibitory rates of mouse hepatoma H22 cell growth in vitro were 31.1% and 30.5%, respectively. The results show that, SEP-S in vitro has a good immunomodulatory effect and anti-tumor effect.
