复方苦参凝胶对豚鼠实验性急性湿疹的作用及机制

目的探讨复方苦参凝胶对豚鼠急性湿疹模型的治疗作用及作用机制。方法健康豚鼠60只随机分为6组,每组10只。除空白对照组外其余各组均采用反复涂抹2,4-二硝基氯苯(DNCB)建造豚鼠急性湿疹模型。造模成功后,空白对照组,模型组,苦参凝胶低、中、高剂量组和阳性对照组豚鼠分别给予空白凝胶,空白凝胶,含苦参碱25、50、100 mg·g~(-1)的复方苦参凝胶和糠酸莫米松凝胶干预,剂量均为0.5 g·kg~(-1),bid。实验第22日观察各组豚鼠右背部皮肤病变并评分,测量皮肤增厚及增重程度。ELISA法测定皮肤组织中TNF-α及IL-1β水平,Western blot法测定皮肤组织中ICAM-1及VCAM-1的表达水平。结果与空白对照组比较,模型组豚鼠皮肤出现肿胀、红斑、糜烂和渗出等症状,皮肤病变评分增高(P<0.01),皮肤明显增厚、增重(P<0.01);苦参凝胶中、高剂量组及阳性对照组上述指标与模型组比较均有不同程度减轻。与空白对照组比较,模型组豚鼠皮肤组织TNF-α及IL-1β含量增加(P<0.01),ICAM-1及VCAM-1的表达增加(P<0.01);与模型组比较,苦参凝胶中、高剂量组及阳性对照组TNF-α及IL-1β含量降低(P<0.01),ICAM-1及VCAM-1的表达下调(P<0.01)。结论复方苦参凝胶对于豚鼠急性湿疹具有明显的疗效,其机制可能与其调节炎症因子、调控相关细胞黏附分子的表达有关。 Objective To investigate the therapeutic effect and mechanism of Compound Kushen Gel on acute guinea pig eczema model. Methods Sixty healthy guinea pigs were randomly divided into 6 groups with 10 rats in each group. Except the blank control group, the guinea pig acute eczema model was constructed by repeatedly applying 2,4-dinitrochlorobenzene (DNCB). After successful modeling, blank control group, model group, low dose group, low dose group, high dose group and positive control group were given blank gel, blank gel, containing matrine 25,50,100 mg · g ~ (-1) Compound Kushen Gel and mometasone furoate gel intervention, the dose was 0.5 g · kg ~ (-1), bid. On the 22nd day of the experiment, the right back and skin lesions of guinea pigs in each group were observed and scored, and the thickening and weight gain of the skin were measured. The levels of TNF-α and IL-1β in skin tissue were measured by ELISA. The expression of ICAM-1 and VCAM-1 in skin tissue were detected by Western blot. Results Compared with the blank control group, the guinea pigs showed swelling, erythema, erosion and exudation in the model group. The skin lesion score increased (P <0.01), the skin thickening and weight gain (P <0.01) Medium and high dose group and positive control group compared with the model group, the above indicators were alleviated to varying degrees. Compared with the blank control group, the content of TNF-α and IL-1β in skin tissue of model group increased (P <0.01) and the expression of ICAM-1 and VCAM-1 increased (P <0.01) The levels of TNF-α and IL-1β in the medium, high dose group and positive control group were decreased (P <0.01), and the expressions of ICAM-1 and VCAM-1 were down-regulated (P <0.01). Conclusion Compound Kushen Gel has obvious curative effect on acute eczema in guinea pigs, which may be related to the regulation of inflammatory factors and the regulation of the expression of cell adhesion molecules.
