目的:探讨抗炎宫乐胶囊治疗血热型崩漏的机制。方法:雌性ICR小鼠按体重随机分为模型组、空白组、抗炎宫乐胶囊高、中、低(1.2,0.6,0.3 g.kg-1)剂量组、宫血宁组、布洛芬组,分别ig给药5,10,10 d。雌性SD大鼠分组情况同小鼠,抗炎宫乐胶囊高、中、低(0.94,0.47,0.24 g.kg-1)剂量组,ig给药20,7,30 d。通过测定抗炎宫乐胶囊对断尾出血时间、血液流变学指标、血小板聚集功能、抗炎镇痛、促性腺激素和性激素调节的影响,研究其治疗血热型崩漏的机制。结果:抗炎宫乐胶囊能够明显缩短小鼠断尾出血时间(P<0.05);缩短大鼠血浆凝血酶原时间(PT)和凝血活酶时间(APTT)(P<0.05),增强二磷酸腺苷(ADP)诱导大鼠血小板聚集功能(P<0.05);减小二甲苯所致小鼠耳肿胀度(P<0.05)和子宫异物埋植所致大鼠子宫肿胀度(P<0.05);延长小鼠疼痛扭体的潜伏期和减少疼痛扭体的次数(P<0.05);提高去卵巢大鼠血清中卵泡刺激素(P<0.05)、黄体生成素(P<0.05)、雌二醇水平(P<0.05)和降低血清中孕酮水平(P<0.05)。结论:抗炎宫乐胶囊对模型动物具有明显的止血、抗炎、镇痛和调节下丘脑-垂体-性腺轴的作用,其治疗血热型崩漏的作用及机制可能与上述环节有关。
Objective: To investigate the mechanism of anti-inflammatory Gong Le Capsule in the treatment of blood-heat-induced metrorrhagia. Methods: The female ICR mice were randomly divided into model group, blank group, anti-inflammatory Gongle capsule high, medium and low (1.2,0.6,0.3 g.kg-1) dose group, Gongsuoning group, ibuprofen Group, respectively, ig administration 5,10,10 d. Female SD rats were grouped with the mice, anti-inflammatory Gong Le capsule high, medium and low doses (0.94,0.47,0.24 g.kg-1) dose group, ig administration 20,7,30 d. The mechanism of anti-inflammatory Gong Le capsule in treating hemorrhagic metrorrhagia was studied by measuring the effects of anti-inflammatory Gong Le capsule on bleeding time, hemorheological parameters, platelet aggregation, anti-inflammatory and analgesic effects, gonadotropin and sex hormone regulation. Results: Kang Yan Gong Le Capsule can significantly reduce the bleeding time of mice (P <0.05), shorten the prothrombin time (PT) and thromboplastin time (APTT) of rats (P <0.05) Adenosine (ADP) induced platelet aggregation in rats (P <0.05), decreased xylene-induced ear swelling in rats (P <0.05) and uterine swelling in rats induced by uterine implantation (P <0.05) (P <0.05), luteinizing hormone (P <0.05), estradiol (P <0.05), prolonging the incubation period of writhing and reducing writhing (P <0.05) (P <0.05) and decreased serum progesterone levels (P <0.05). Conclusion: Kangyan Gongle Capsule has obvious hemostatic, anti-inflammatory, analgesic and hypothalamic-pituitary-gonadal axis effects on model animals. The effect and mechanism of anti-inflammatory Gonglele capsule on heat-induced metrorrhagia may be related to the above aspects.
