目的考察氧化槐果碱(OSC)抑制SW480细胞及诱导其产生凋亡的作用。方法根据用药剂量分组,用MTT法测定氧化槐果碱对SW480细胞增殖的影响,荧光双染观察细胞形态变化,计数凋亡细胞比率;定时定量聚合酶链式反应(real-time PCR)检测凋亡基因caspase-3和Bcl-2的变化。结果 OSC对体外培养的SW480细胞增殖具有明显抑制作用;OSC作用后凋亡细胞比例显著增加;凋亡基因caspase-3 mRNA表达上调而Bcl-2 mRNA表达下调。结论OSC可抑制体外培养的SW480细胞生长增殖,其作用机制与阻滞细胞周期进程、诱导细胞凋亡有关。
Objective To investigate the effect of OSC on SW480 cells and induce their apoptosis. Methods According to the dose of treatment group, the effect of SAC on the proliferation of SW480 cells was determined by MTT assay. The changes of cell morphology were observed by fluorescence double staining and the ratio of apoptotic cells was counted. The expression of apoptotic cells was detected by real-time polymerase chain reaction The changes of death gene caspase-3 and Bcl-2. Results OSC significantly inhibited the proliferation of SW480 cells in vitro. The proportion of apoptotic cells increased significantly after OSC treatment. The expression of caspase-3 mRNA was up-regulated but the expression of Bcl-2 mRNA was down-regulated. Conclusion OSC can inhibit the growth and proliferation of SW480 cells cultured in vitro, and its mechanism is related to arresting cell cycle progression and inducing apoptosis.
