目的观察孕酮、左炔诺孕酮和醋酸甲羟孕酮对体外培养子宫肌瘤细胞bcl-2基因表达的影响。方法体外培养子宫肌瘤细胞,并采用半定量RT-PCR法检测不同浓度孕酮、左炔诺孕酮和醋酸甲羟孕酮干预后子宫肌瘤细胞bcl-2凋亡抑制基因的表达。结果成功进行子宫肌瘤细胞的原代培养,加入浓度为0.1、10、1000 ng/ml孕酮、左炔诺孕酮和醋酸甲羟孕酮后,均上调子宫肌瘤细胞内bcl-2的表达,与对照组相比,差异有统计学意义(P<0.05),并且呈剂量依赖性。结论孕酮和左炔诺孕酮和醋酸甲羟孕酮可增加体外培养子宫肌瘤细胞bcl-2基因的表达。
Objective To observe the effect of progesterone, levonorgestrel and medroxyprogesterone acetate on bcl-2 gene expression in uterine fibroids in vitro. Methods Uterine fibroids were cultured in vitro and the expression of bcl-2 mRNA was detected by semi-quantitative RT-PCR in different concentrations of progesterone, levonorgestrel and medroxyprogesterone acetate. Results The primary culture of uterine leiomyoma cells was successfully carried out.
