建立了一种基于硼酸亲和作用的pH指示剂比色法,并应用于糖和核苷类样品的含量测定。选取苯硼酸为硼酸试剂,以糖类和核苷为测试对象,考察并比较不同波长下采用甲基橙和百里酚兰指示剂时溶液浓度与吸光度变化值之间的线性关系。结果表明,糖类物质浓度在10~300 mmol/L范围内,核苷类物质浓度在2~36 mmol/L范围内,采用百里酚兰指示剂于600 nm下测定,线性关系较好。验证实验结果表明方法具有较好的稳定性,准确度和精密度。
A pH indicator colorimetric method based on the affinity of boric acid was established and applied to the determination of sugar and nucleoside samples. Phenylboronic acid was selected as a boric acid reagent, and carbohydrates and nucleosides were tested. The linear relationship between the concentration of the solution and the change of the absorbance under different wavelengths using methyl orange and thymol blue indicator was investigated. The results showed that the concentration of saccharide was in the range of 10 ~ 300 mmol / L, the nucleoside concentration was in the range of 2 ~ 36 mmol / L, and the calibration curve was good at 600 nm with thymol blue indicator. The experimental results show that the method has good stability, accuracy and precision.
