从灵芝 (Ganodermalucidum)加硒深层培养的菌丝中 ,分别经水、碱水提取 ,醇析 ,透析 ,脱色 ,脱蛋白 ,DEAE—Cellulose柱层析和SepharoseCL4B柱层析纯化后 ,得SeGLP - 2A。经SephadexG - 10 0、聚丙烯酰胺凝胶电泳和紫外光谱分析鉴定 ,SeGLP - 2A为均一级分。GC与TLC分析表明 :SeGLP - 2A含有 4种单糖Gal、Xyl、Glu、Rha ,其单糖摩尔比为n (Gal) :n (Xyl) :n (Glu) :n (Rha) =2 5 5 9∶16 30∶1 0 0∶0 86。经红外光谱分析 ,确定SeGLP - 2是由α -糖苷键连接的吡喃多糖
Sepharose-2A was obtained from the hyphae (Ganodermalucidum) supplemented with selenium in a deep-cultured hyphae, purified by water, aqueous alkali, alcohol extraction, dialysis, decolorization, deproteinization, DEAE-Cellulose column chromatography, and Sepharose CL4B column chromatography. . Sephadex G-10, polyacrylamide gel electrophoresis, and UV spectroscopic analysis identified SeGLP-2A as a homogeneous fraction. GC and TLC analysis showed that SeGLP-2A contains 4 monosaccharides: Gal, Xyl, Glu, and Rha. The molar ratio of monosaccharides is n (Gal):n (Xyl):n (Glu):n (Rha) =2 5 5 9:16 30:10 0:0 86. By infrared spectroscopy, it was determined that SeGLP-2 is a pyran polysaccharide linked by α-glycosidic bonds.
