目的 :探讨 NHL 患者骨髓细胞 DNA倍体类型与细胞动力学参数之间的关系。方法 :采用流式细胞术对尚未进行化疗的 32 7例 NHL 患者骨髓细胞 DNA含量进行了检测 ,并分析DNA倍体类型与细胞凋亡 (Apo)水平及 S-期细胞比率 (SPF)的关系。结果 :NHL 患者骨髓细胞 DNA倍体类型分为二倍体 (D,136例 ) ,异倍体 (H,191例 )。患者骨髓细胞 Apo检出率显著低于正常对照组 (P<0 .0 1) ,而 SPF比率却显著高于正常对照组 (P<0 .0 1)。二倍体 NHL 患者骨髓 Apo检出率显著高于异倍体肿瘤组织 (P<0 .0 1) ,SPF比率显著低于异倍体组织 (P<0 .0 1)。在 NHL 患者骨髓细胞中 ,含有不同数量异倍体克隆者的 Apo检出率和 SPF比率均有显著性差异 (P<0 .0 1)。 SPF升高者的骨髓细胞的 Apo和 DNA异倍体检出率均显著低于 SPF降低者 (P<0 .0 5 )。结论 :NHL 能导致患者骨髓细胞 Apo水平显著降低 ,细胞增生活性显著增加。而且这些变化与肿瘤组织倍体类型和 DNA异倍体克隆数均有十分密切的关系
Objective: To investigate the relationship between DNA ploidy and cell kinetic parameters of bone marrow cells in NHL patients. Methods: The DNA content of bone marrow cells in 32 7 NHL patients who had not been treated with chemotherapy was detected by flow cytometry. The relationship between DNA ploidy type and apoptosis rate (Apo) and S phase ratio (SPF) . Results: The DNA ploidy types of NHL patients were divided into diploid (D, 136 cases) and aneuploid (H, 191 cases). The detection rate of Apo in patients with bone marrow cells was significantly lower than that in the normal control group (P <0.01), but the SPF rate was significantly higher than that in the normal control group (P <0.01). The detection rate of bone marrow Apo in diploid NHL patients was significantly higher than that in aneuploid tumor tissues (P <0.01), and the SPF rate was significantly lower than aneuploid tissues (P <0.01). In NHL patients with bone marrow cells, aneuploid clones with different numbers of Apo detection rate and SPF ratio were significantly different (P <0.01). The detection rates of Apo and DNA aneuploidy in bone marrow cells with elevated SPF were significantly lower than those with decreased SPF (P <0.05). CONCLUSIONS: NHL can lead to a significant reduction of Apo levels and a significant increase of cell proliferation in bone marrow cells of patients. And these changes are closely related to the type of tumor tissue and the number of cloned aneuploid cells
