采用国产C1 8反相柱 ,研究了异硫氰酸苯酯 (PITC)柱前衍生高效液相色谱法测定复合氨基酸中牛磺酸的可行性 ,测定了淡水珍珠中牛磺酸的含量 ,结果表明 ,采用甲醇流动相体系 ,在适宜的色谱条件下 ,牛磺酸可获得较好的分离 ,并且在 8~ 4 0 μg/mL浓度范围内与峰面积成良好的线性关系 ,峰面积定量的相对标准偏差为 0 .7% (n =5) ,牛磺酸的PITC衍生物溶液室温存放 3 6h不变质 ,用本法测得淡水珍珠样品中牛磺酸含量为 0 .0 2 %。
The feasibility of determination of taurine in compound amino acids by PITC pre-column derivatization with high performance liquid chromatography (HPLC) was studied using a domestic C1 8 reversed-phase column. The content of taurine in freshwater pearls was determined. Results The results showed that taurine could be separated well under the suitable chromatographic conditions by methanol mobile phase system and had a good linear relationship with the peak area in the concentration range of 8 ~ 40 μg / mL. The peak area was The relative standard deviation was 0.7% (n = 5). The solution of taurine PITC derivative was not deteriorated for 36h at room temperature. The content of taurine in freshwater pearl sample was 0.2% by this method.
