应用改进的免疫电镜技术,对不同戊型肝炎病毒株(87A,G-93-2,G93-4和F86)的形态进行了观察。用本实验室制备的抗-HEV(87A株)单克隆抗体(mAb),与2BS细胞培养,并加入经PEG沉淀浓集的病毒悬液温育后制样。结果可见:国内不同流行区分离的暴发和散发的3株HEV颗粒呈聚集状态,毒粒有空心、实心两种形态,直径25nm~36nm,平均30.6nm。病毒的形态大小及聚集态势与戊肝患者粪便悬液及实验感染猴标本所见十分相近;免疫电镜的结果与mAb所做的微量中和及血凝抑制试验结果相一致,也与多克隆抗体(pAb)的研究结果可比较。此方法简便实用,适合滴度低或纯化有困难的病毒形态学研究。
The morphological changes of different hepatitis E virus strains (87A, G-93-2, G93-4 and F86) were observed using improved immunoelectron microscopy. The anti-HEV (87A strain) monoclonal antibody (mAb) prepared in our laboratory was incubated with 2BS cells and incubated with the virus suspension concentrated by PEG precipitation. The results showed that the three outbreaks and exudes of HEV particles isolated in different endemic areas in China were aggregated. The virion was hollow and solid with diameters of 25 nm-36 nm with an average of 30.6 nm. Morphology of the virus and its aggregation pattern were similar to those of the eosinophils of Hepatitis E patients and the infected monkeys. The results of immunoelectron microscopy were in good agreement with the results of the microneutralization and hemagglutination inhibition test by mAbs, (PAb) study results can be compared. This method is simple and practical, suitable for low virus titer or purification of morphological studies.
