建立RP-HPLC法同时测定芫花及其醋制品中两种香豆素类成分含量测定的方法采用反相高效液相色谱法,采用AgiLent EcLipse XDB-C18色谱柱;流动相条件为乙腈(A)-5%冰乙酸(B),进行梯度洗脱;检测波长为:327 nm;流速:1.0 mL/min;柱温:25℃。瑞香素和伞形花内酯在各自线性范围内线性关系良好,10个来源的芫花药材及醋制品中两种香豆素类成分的含量有一定差异。本法灵敏度高,重现性好、所测结果稳定,可用于芫花药材及其炮制品的质量控制。
To establish an RP-HPLC method for the simultaneous determination of two coumarins in Daphne genkwa and its vinegar by reversed-phase high performance liquid chromatography with AgiLent EcLipse XDB-C18 column. The mobile phase consisted of acetonitrile (A ) -5% glacial acetic acid (B), gradient elution; detection wavelength: 327 nm; flow rate: 1.0 mL / min; Daphnetin and umbelliferone had good linearity in their respective linear ranges. The contents of two coumarins in 10 samples of Daphne genkwa and its vinegar products were different. The method of high sensitivity, good reproducibility, the measured results are stable, can be used for quality control of medicines and their processed products.
