目的制备胸腺五肽多囊脂质体,并考察其包封率和体外释放情况。方法采用复乳法制备胸腺五肽多囊脂质体,并用单因素、正交实验对处方进行优化。结果制备的胸腺五肽多囊脂质体的外观圆整均匀,形态良好,包封率为80.15%±3%。体外释放的零级动力学方程为:Y=0.536X+20.431(r2=0.9814),132 h的累积释放率为92.01%。结论复乳法制备的胸腺五肽多囊脂质体的工艺可行,包封率较高,有明显的缓释特征。
Objective To prepare thymopentin multilocularis liposomes and investigate their encapsulation efficiency and in vitro release. Methods Twosome milk liposomes were prepared by double emulsion method. The prescription was optimized by single factor and orthogonal experiment. Results The appearance of thymopentin multivesicular liposomes was round and uniform and the morphology was good. The entrapment efficiency was 80.15% ± 3%. The zero-order kinetic equation for in vitro release was: Y = 0.536X + 20.431 (r2 = 0.9814), and the cumulative release rate at 132 h was 92.01%. Conclusion The preparation of thymopentin multilocular liposomes by double emulsion method has the advantages of high encapsulation efficiency and obvious sustained release characteristics.
