目的探讨魔芋多糖(konjac polysaccharide,KP)对小鼠肠道吸收功能的抑制作用与机制。方法将实验动物分为正常对照(N)、高脂(HF)和KP高、中、低剂量(KPH、KPM、KPL)共5个组,连续喂饲20d。紫外分光光度法测定小肠粘膜Na+-K+-ATP酶活性,血糖仪测定血糖并称量体重和粪便湿、干重等。结果HF组和KPH组的Na+-K+-ATP酶活性分别为16.2±1.48和11.2±1.10μmolPi/(mgpro·h),体重和餐后血糖分别为34.3±2.07g、7.5±1.15mmol/L和28.1±1.95g、4.8±0.73mmol/L。两组间各项指标的差别均有显著性意义(P<0.05)。结论KP降低小鼠餐后血糖和血清胆固醇水平,抑制体重增长、降低小肠粘膜Na+-K+-ATP酶活性,对肠道吸收功能有抑制作用。
Objective To investigate the inhibitory effect and mechanism of konjac polysaccharide (KP) on intestinal absorption in mice. Methods The experimental animals were divided into 5 groups: normal control (N), high fat (HF) and KP high, medium and low dose (KPH, KPM, KPL) for 20 days. The activity of Na + -K + -ATPase in small intestine mucosa was measured by ultraviolet spectrophotometry. The blood glucose was measured by blood glucose meter and the body weight and feces wet weight and dry weight were measured. Results The activities of Na + -K + -ATPase in HF and KPH groups were 16.2 ± 1.48 and 11.2 ± 1.10μmolPi / (mgpro · h) respectively. The body weight and postprandial blood glucose were 34.3 ± 2.07g and 7.5 ± 1.15mmol / L and 28.1 ± 1.95g, 4.8 ± 0.73mmol / L. The differences between the two groups were significant (P <0.05). Conclusion KP can reduce postprandial blood glucose and serum cholesterol levels, inhibit the growth of body weight, decrease the activity of Na + -K + -ATPase in intestinal mucosa, and inhibit the intestinal absorption.
