目的 研究坛紫菜中酸性多糖的化学结构。方法 采用CTAB结合法、超滤法使酸性多糖PP1与中性多糖PP2分离 ,DEAE -纤维素、琼脂糖凝胶使PP1纯化、紫外光谱、红外光谱、气相色谱、高碘酸氧化、Smith降解等色谱与化学方法测定PP1化学结构。结果 PP1由半乳糖、3- 6 -内醚 -L -半乳糖、甘露糖、葡萄糖组成 ,具α ,β型糖苷键 ,连接方式有 ( 1→ 3)、( 1→ 4)、( 1→ 2 ) 3种、其总糖、3,6 -内醚 -L -半乳糖及硫酸基的含量分别为 85 .41%、5 .95 %、14.0 %。PP1分子量约为 47.9万。结论首次对坛紫菜酸性多糖的化学结构作了较为细致的研究
Objective To study the chemical structure of acid polysaccharides in Porphyra haitanensis. Methods The acid polysaccharide PP1 and the neutral polysaccharide PP2 were separated by CTAB binding method and ultrafiltration method. The DEAE-cellulose and agarose gels were used for the purification of PP1, ultraviolet spectroscopy, infrared spectroscopy, gas chromatography, periodate oxidation, Smith degradation, etc. Chromatography and chemical methods were used to determine the chemical structure of PP1. Results PP1 consisted of galactose, 3- 6 -endoether-L-galactose, mannose, and glucose, with α,β glycosidic bonds. The connection methods were (1→ 3), (1→ 4), (1→ 2) The content of 3 kinds of total sugars, 3,6-lactone-L-galactose and sulfate groups were 85.41%, 5.95%, and 14.0%, respectively. PP1 has a molecular weight of about 479,000. Conclusion For the first time, the chemical structure of acid polysaccharide from Porphyra haitanensis was studied in detail.
