目的探讨 CD_(15) mRNA 及其蛋白表达与胃癌病理生物学行为的关系及 CD_(15) mRNA 表达与其蛋白表达之间的相关性,为观测胃癌侵袭转移潜能和评估胃癌患者预后寻求一个新的客观生物学指标。方法应用原位杂交及高敏感性 CSA 免疫组化方法和 CD_15s特异性 mAb,对于手术切除的17例早期胃癌,21例中期胃癌和57例晚期胃癌组织进行 CD_15mRNA 及其蛋白检测,并结合肿瘤的病理生物学行为和临床随访资料进行分析。结果在原发性胃癌中,CD_(15) mRNA 及其蛋白的表达阳性率分别为89.5%(85/95)和86.3%(82/95).CD_(15)mRNA 及其蛋白表达阳性率均在晚期胃癌96.5%(55/57),93.O%(53/57)明显高于早期胃癌76.5%(13/17),70.6%(12/17)和中期胃癌81.0%(17/21),P<0.05.CD_(15) mRNA 表达与蛋白表达水平具有一致性,均与胃癌浆膜浸润,淋巴结转移和患者预后呈正相关(P<0.05).15例原发和淋巴结转移性胃癌组织均呈CD_(15) mRNA 及其蛋白阳性表达。结论 CD_(15) mRNA 及其蛋白异常表达与胃癌的临床病理生物学行为密切相关,特别是与胃癌细胞的转移潜能和胃癌患者的不良预后密切相关,CD_(15)蛋白水平的表达可以间接反映其mRNA 转录水平,并可作为是预测胃癌转移潜能和评估胃癌患者预后的一个新的生物学指标。
