采用兔自身对照交叉法,给予自制盐酸左氧氟沙星滴眼液,并以两种市售滴眼液作参比制剂,采用HPLC法测定给药后兔眼房水中的左氧氟沙星,自制盐酸左氧氟沙星滴眼液和两种参比制剂的主要药动学参数为:AUC_(0-∞)(5.10±0.46)、(2.48±0.32)和(4.74±0.10)μg·ml~(-1)·h,t_(1/2)(1.50±0.50)、(2.26±0.48)和(1.91±0.26)h,c_(max)(1.53±0.59)、(1.12±0.11)和(1.49±0.16)μg/ml。自制滴眼液与两种市售滴眼液的相对生物利用度为213.2%和110.4%。
Levofloxacin Hydrochloride Eye Drops were given by self-made crossover method. Two commercially available eye drops were used as reference preparations. HPLC was used to measure levofloxacin in rabbits aqueous humor and self-made levofloxacin eye drops The main pharmacokinetic parameters of the two reference preparations were as follows: AUC 0 -∞ (5.10 ± 0.46), (2.48 ± 0.32) and (4.74 ± 0.10) μg · ml -1 · h, t 1 /2)(1.50 ± 0.50), (2.26 ± 0.48) and (1.91 ± 0.26) h, c max (1.53 ± 0.59), (1.12 ± 0.11) and (1.49 ± 0.16) μg / ml, respectively. The relative bioavailability of homemade eye drops versus two marketed eye drops was 213.2% and 110.4%.
