目的:观察血管内皮生长因子(VEGF)受体flk-1在不典型增生及鳞癌中的表达,用图象分析系统定量的方法观察flk-1与喉癌的生长、浸润及转移的关系。方法:用抗flk-1抗体,在经过微波处理组织暴露抗原后,进行标准化的EliteABC染色,并通过图象分析系统确定flk-1在不典型增生、鳞癌中的表达。结果:在不典型增生及鳞癌中flk-1均有表达。在组织中,flk-1不仅在血管内皮细胞中、在部分浆细胞和巨噬细胞中也呈阳性反应。鳞癌组中的flk-1的表达明显高于不典型增生组(P<0.05)。在鳞癌中,随着分化程度的下降,flk-1表达有逐渐递增的趋势。在鳞癌组中发生转移的病变flk-1的表达明显高于未转移组(P<0.05)。相关分析显示flk-1的表达与肿瘤的临床分期密切相关。结论:VEGF通过自分泌和旁分泌的途径影响肿瘤细胞生长及间质中新生血管的形成,肿瘤细胞的生长与血管的发生相互依赖。flk-1与喉癌的生长、浸润及转移密切相关,flk-1可能作为一个判断预后的重要指标。
OBJECTIVE: To observe the expression of vascular endothelial growth factor (VEGF) receptor flk-1 in atypical hyperplasia and squamous cell carcinoma, and to observe the relationship between flk-1 and growth, invasion, and metastasis of laryngeal carcinoma by image analysis system. METHODS: Anti-flk-1 antibody was used to perform standardization of Elite ABC staining on exposed tissues of microwave-treated tissues and the expression of flk-1 in dysplasia and squamous cell carcinoma was determined by image analysis system. RESULTS: Flk-1 was expressed in atypical hyperplasia and squamous cell carcinoma. In tissues, flk-1 is also positive in not only vascular endothelial cells but also some plasma cells and macrophages. The expression of flk-1 in the squamous cell carcinoma group was significantly higher than that in the atypical hyperplasia group (P<0.05). In squamous cell carcinoma, the expression of flk-1 is gradually increasing as the degree of differentiation decreases. The expression of flk-1 in the squamous cell carcinoma group was significantly higher than that in the non-metastatic group (P<0.05). Correlation analysis showed that the expression of flk-1 is closely related to the clinical stage of the tumor. CONCLUSION: VEGF affects the growth of tumor cells and the formation of new blood vessels in the interstitium through autocrine and paracrine pathways. The growth of tumor cells is dependent on the occurrence of blood vessels. Flk-1 is closely related to the growth, invasion and metastasis of laryngeal carcinoma, and flk-1 may be used as an important indicator of prognosis.
