目的:建立同时测定难治性癫痫(IE)患者脑组织中拉莫三嗪(LTG)、苯巴比妥(PB)、卡马西平(CBZ)及苯妥英钠(PHT)药物浓度的HPLC法。方法:流动相为10mmol·L~(-1)磷酸二氢钾缓冲液:甲醇:乙腈:丙酮=55:22:12:11,流速1ml·min~(-1),柱温:室温,检测波长:210 nm,采用外标法。结果:LTG、CBZ浓度在0.5~40μg·ml~(-1)和PB、PHT浓度在1~80μg·ml~(-1)范围内,被测药物峰面积与浓度呈良好的线性关系,相对回收率均高于85%,日内、日间RSD均小于12%。结论:本法操作简便、准确度高,可用于测定难治性癫痫患者脑组织中4种抗癫痫药物浓度。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of lamotrigine (LTG), phenobarbital (PB), carbamazepine (CBZ) and phenytoin sodium (PHT) in patients with refractory epilepsy (IE) Methods: The mobile phase consisted of 10 mmol·L -1 potassium dihydrogen phosphate buffer: methanol: acetonitrile: acetone = 55:22:12:11, the flow rate was 1 ml · min -1. The column temperature was measured at room temperature Wavelength: 210 nm, using external standard method. Results: The concentration of LTG and CBZ was in the range of 0.5-40 μg · ml -1 and the concentration of PB and PHT was in the range of 1-80 μg · ml -1. The peak area and the concentration of the tested drug showed a good linear relationship, Recoveries were higher than 85%, day, day RSD were less than 12%. Conclusion: This method is simple, accurate and can be used to determine the concentration of 4 antiepileptic drugs in brain tissue of patients with refractory epilepsy.
