目的探讨氯沙坦对1型糖尿病大鼠心肌组织中白细胞介素-18(IL-18)和核因子-κB p65(NF-κB p65)表达的影响及其对糖尿病心肌病的作用。方法雄性SD大鼠54只,随机分为正常对照组(NC组);糖尿病组(DM组);氯沙坦干预组(DM-L组)。于实验的4周、8周和12周,测定血糖、血清胰岛素、心重指数及心肌细胞面积,光镜下观察心肌结构形态变化,免疫组化测定心肌IL-18和NF-κB p65的蛋白变化,于实验12周用荧光定量RT-PCR测定心肌中IL-18和NF-κB p65的mRNA水平。结果与正常对照组相比,糖尿病组和氯沙坦干预组心重指数、心肌细胞面积、心肌组织IL-18和NF-κB p65的蛋白及mRNA表达均显著增高(P<0.05),且氯沙坦干预组上述指标显著低于糖尿病组(P<0.05)。结论糖尿病心肌组织中IL-18和NF-κB p65表达升高与糖尿病心肌病变的发生密切相关,氯沙坦能抑制糖尿病大鼠心肌IL-18和NF-κB p65表达,减轻心肌病变,从而对糖尿病心肌病起到保护作用。
Objective To investigate the effect of losartan on the expression of interleukin-18 (IL-18) and nuclear factor-κB p65 (NF-κB p65) in myocardium of type 1 diabetic rats and its effect on diabetic cardiomyopathy. Methods Fifty-four male SD rats were randomly divided into normal control group (NC group), diabetic group (DM group) and losartan intervention group (DM-L group). Blood glucose, serum insulin, cardiac index and myocardial cell area were measured at 4 weeks, 8 weeks and 12 weeks after the experiment. The morphological changes of myocardium were observed under light microscope. The protein expressions of myocardial IL-18 and NF-κB p65 were measured by immunohistochemistry The mRNA levels of IL-18 and NF-κB p65 in myocardium were determined by fluorescence quantitative RT-PCR at 12 weeks of experiment. Results Compared with the normal control group, the protein and mRNA expressions of IL-18 and NF-κB p65 in the diabetic group and losartan group were significantly increased (P <0.05) The above indicators in the intervention group were significantly lower than those in the diabetic group (P <0.05). CONCLUSION: The increased expression of IL-18 and NF-κB p65 in diabetic myocardium is closely related to the development of diabetic cardiomyopathy. Losartan can inhibit the expression of IL-18 and NF-κB p65 in diabetic rats and relieve the cardiomyopathy, Diabetic cardiomyopathy plays a protective role.
