目的观察心肌细胞在血管紧张素Ⅱ(AngⅡ)作用后反映能量的酶与脂质过氧化反应的变化及不同类中药对其的影响。方法用AngⅡ作用于培养的SD大鼠乳鼠心肌细胞,观察作用24、48、72、96 h细胞上清液中乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)的变化及益气、活血、益气活血中药的作用。结果在AngⅡ作用48 h后,细胞上清液中LDH、SDH明显增加(P<0.01),MDA含量明显升高,SOD活性明显降低(P<0.01),各中药组LDH、SDH明显降低,且可明显减少MDA含量、升高SOD活性(P<0.05或P<0.01)。结论AngⅡ作用一定时间后,伴随着能量的变化和引发的脂质过氧化反应,使心肌细胞的功能受到影响,益气、活血、益气活血中药均有保护作用,尤以益气活血药作用明显。
Objective To observe the changes of enzymes and lipid peroxidation that reflect the energy of angiotensin II (Ang II) in cardiomyocytes and the effects of different Chinese herbs on it. Methods AngII was used to treat cultured SD rat cardiomyocytes, and lactate dehydrogenase (LDH), succinate dehydrogenase (SDH), malondialdehyde were observed in cell supernatants at 24, 48, 72, and 96 h. (MDA), superoxide dismutase (SOD) changes and the role of Qi, blood circulation, Qi and activating blood circulation of traditional Chinese medicine. Results After treatment with AngII for 48 h, the LDH and SDH in the supernatant of the cells increased significantly (P<0.01), the MDA content increased significantly, and the SOD activity decreased significantly (P<0.01). The LDH and SDH of the traditional Chinese medicine group were significantly decreased. MDA content and SOD activity were significantly decreased (P<0.05 or P<0.01). Conclusion After a certain period of AngII action, accompanied by changes in energy and induced lipid peroxidation, the function of cardiomyocytes is affected. Yiqi, Huoxue and Yiqihuoxue herbs all have protective effects, especially the effect of qi and blood circulation drugs. obvious.
