目的研究犬肺癌模型抑癌基因表达与支气管动脉造影表现的相关性,探讨肺癌早期诊断的新方法、新标准。材料与方法22只犬,经气管插管由同轴微导管定位向右肺膈叶灌注甲基胆蒽、二乙基亚硝胺和超液化碘油混悬液,诱导犬肺癌模型,在不同时期进行基因检测和影像、病理学检查。结果p53基因蛋白随诱导时间延长,阳性表达率逐渐升高,p16逐渐减低。CD31血管内皮标记表达阳性与支气管动脉造影所见肿瘤样血管及染色吻合。结论CD31血管内皮细胞标记阳性表达和支气管动脉造影发现的肿瘤血管及染色是肺癌早期诊断的参考指标;早期检测p53基因突变与p16基因失活对提高早期诊断率和5年治愈率有重要意义。
Objective To study the relationship between tumor suppressor gene expression and bronchial arteriography in canine lung cancer model and to explore a new method and new standard for early diagnosis of lung cancer. MATERIALS AND METHODS Twenty-two dogs were anesthetized with different concentrations of methylcholanthrene, diethylnitrosamine and lipiodol in suspension in the right lung by intraperitoneal microcatheter via endotracheal intubation. Period of genetic testing and imaging, pathological examination. Results p53 gene protein with induction time, the positive expression rate gradually increased, p16 decreased. The positive expression of CD31 vascular endothelial marker was consistent with the tumor-like vessels and staining seen in bronchial artery angiography. Conclusion The positive expression of CD31 vascular endothelial cells and tumor vessels and staining found by bronchial artery angiography are the reference indexes for the early diagnosis of lung cancer. The early detection of p53 gene mutation and inactivation of p16 gene are of great significance to improve the early diagnosis rate and 5-year cure rate.
