采用乳化超声法制备奥沙利铂纳米结构脂质载体(oxaliplatin-loaded nanostuctured lipid carriers,OP-NLC),通过正交试验筛选出最优处方。运用激光粒度仪、zeta电位仪、透射电镜(transmission electron microscope,TEM)、差示扫描量热法(differential scanning calorimetry,DSC)、X衍射(X-ray diffraction,XRD)以及高效液相色谱法(high performance liquid chromatography,HPLC)等考察其理化性质。结果显示,优化条件下制备的OP-NLC多为类球形粒子,平均粒径(111±20)nm,zeta电位为(?27.4±13.1)mV,包封率(encapsulation efficiency,EE)为(77.4±2.5)%,药物含量为(0.8±1.5)mg·mL?1,DSC及XRD表明药物以非结晶状分散于纳米粒中。体外释放实验表明,OP-NLC在pH4.5的缓冲液中释药前期有突释现象,后期则具有明显的缓释特征。
The oxaliplatin-loaded nanostuctured lipid carriers (OP-NLC) were prepared by emulsified ultrasound, and the optimal prescription was screened by orthogonal test. The effects of laser scanning electron microscopy, laser scanning electron microscopy, transmission electron microscopy (TEM), differential scanning calorimetry (DSC), X-ray diffraction (XRD) and high performance liquid chromatography high performance liquid chromatography, HPLC) and other physical and chemical properties. The results showed that OP-NLC prepared under optimal conditions was mostly spherical and had an average particle size of (111 ± 20) nm and a zeta potential of (-27.4 ± 13.1) mV with an encapsulation efficiency (EE) of (77.4 ± 2.5)%, and the drug content was (0.8 ± 1.5) mg · mL -1. DSC and XRD indicated that the drug was dispersed in the nanoparticles in a non-crystalline state. In vitro release experiments showed that OP-NLC in the pH4.5 buffer release early burst phenomenon, the latter has a significant slow release characteristics.
