分别采用Alcalase酶与酸性蛋白酶、Alcalase酶与胃蛋白酶两种组合方式酶解葵花籽分离蛋白,依次得到多肽A和多肽B两组酶解产物。研究表明:这两组酶解产物对.OH、O2-.和DPPH.自由基的清除作用均明显高于葵花籽分离蛋白(P<0.05);与谷胱甘肽(GSH)相比,多肽A在一定范围内清除.OH和O2-.的效果显著高于GSH(P<0.05),清除DPPH.的效果低于GSH;而多肽B清除O2.-和DPPH.的效果不及GSH。电泳技术和凝胶层析分析发现,多肽A、B两组酶解产物的相对分子质量分布有所不同,尽管这两种多肽的分子质量大部分都小于3 000 u,但是多肽A中小于1 000 u的多肽比例大于多肽B,达到62.44%。
Respectively, Alcalase enzyme and acid protease, Alcalase enzyme and pepsin were used to separate the sunflower seed protein from the sunflower seeds, then the two hydrolysates of polypeptide A and polypeptide B were obtained. The results showed that the scavenging effects of the two hydrolyzate groups on the free radicals of OH, O2- and DPPH were significantly higher than that of the sunflower seed protein (P <0.05). Compared with the GSH, A was scavenged within a certain range. The effect of OH and O2- was significantly higher than that of GSH (P <0.05), and the effect of DPPH was less than that of GSH. However, the effect of peptide B on O2.- and DPPH removal was less than that of GSH. Electrophoresis and gel analysis showed that the relative molecular mass distributions of digested peptides A and B were different. Although most of the molecular weights of these two peptides were less than 3 000 u, The proportion of polypeptide with 000 u was larger than polypeptide B, reaching 62.44%.
