本文利用器官培养的方法,观察长管状软骨的分化,成熟和化骨过程。软骨的生长在第14天长径生长最迅速,此后渐趋停止。电镜观察,培养2—4天的软骨细胞核与胞浆的比值大,细胞内可见多数小泡体。培养6天时原始骨化中心开始出现,粗面内质网和线粒体丰富,细胞开始分泌基质,基质内有具单位膜的基质小泡形成。说明化骨准备工作已经完善。第14天时肥大层软骨细胞内粗面内质网和线粒体更加丰富,内质网扩张呈池状,其内含有合成的点状或丝状物。本文因此提出利用软骨器官培养方法,在培养8—10天时,施加实验因素最宜。
In this paper, the method of organ culture was used to observe the differentiation, maturation and bone formation of long tubular cartilage. On the 14th day, the growth of cartilage grew most rapidly and then gradually stopped. Electron microscopy, cultured 2-4 days of chondrocytes nuclear and cytoplasm ratio, the majority of vesicles can be seen in cells. The original ossification center began to appear when cultured for 6 days, and the rough endoplasmic reticulum and mitochondria were abundant. The cells started to secrete the stroma, and the stroma had the stroma vesicles formed. Description of bone preparation has been completed. On the 14th day, the rough endoplasmic reticulum and mitochondria were more abundant in the hypertrophic layer of chondrocytes, the endoplasmic reticulum dilated in a pool, which contained synthetic dots or filaments. This paper therefore proposed the use of cartilage organ culture methods, the training of 8-10 days, the most appropriate experimental factors.
