目的研究2-ME联合抗坏血酸在诱导白血病细胞凋亡中的作用以及相关作用机制。方法用显微镜观察K562细胞凋亡情况;通过流式细胞术分析凋亡率;通过EMSA检测NF-κB的活性。结果 2μmol/L2-ME+62.5μmol/LAA组可诱导K562细胞凋亡。2-ME+AA以剂量依赖性方式诱导凋亡,在浓度(2~8μmol/L)时凋亡呈时间-剂量依赖。2-ME可诱导NF-κBDNA结合活性以剂量依赖性方式降低。结论 2-ME联合抗坏血酸可诱导白血病细胞以时间-剂量依赖式凋亡,其诱导凋亡的机制与NF-κB通路有关。
Objective To study the role of 2-ME combined with ascorbic acid in inducing leukemia cell apoptosis and its related mechanism. Methods The apoptosis of K562 cells was observed by microscope. The apoptosis rate was analyzed by flow cytometry. The activity of NF-κB was detected by EMSA. Results The apoptosis of K562 cells was induced by 2μmol / L 2-ME + 62.5μmol / LAA. 2-ME + AA induced apoptosis in a dose-dependent manner, and the apoptosis was dose-dependent at the concentration of 2 ~ 8μmol / L. 2-ME induced a decrease in NF-κB DNA binding activity in a dose-dependent manner. Conclusions 2-ME combined with ascorbic acid can induce the apoptosis of leukemia cells in a time-and dose-dependent manner. The mechanism of apoptosis induced by 2-ME is related to NF-κB pathway.
