采用包被多克隆抗体直接竞争ELISA法研制一种检测水样、蔬菜和粮食样品中三唑磷残留的酶联免疫吸附测定法(ELISA)试剂盒。将不同浓度的三唑磷添加到水样、蔬菜和粮食样品中,用制备的试剂盒检测三唑磷残留并用气相色谱分析法(GC)进行验证,结果样品的加标回收率在65%~125%之间,变异系数<15%。对试剂盒的灵敏度、特异性、精密度、准确度和有效期进行测定,结果表明:研制的试剂盒最低检测限为4.13ng/mL,线性检测范围3.9~500ng/mL;在4℃或-20℃条件下试剂盒有效期可达6个月。用ELISA试剂盒检测三唑磷残留具有可批量分析样品,快速、简便、灵敏、经济的特点,可广泛应用于环境监测和安全食品的生产。
A polyclonal antibody direct competitive ELISA was developed to develop an enzyme linked immunosorbent assay (ELISA) kit for the determination of triazophos in water samples, vegetables and food samples. Different concentrations of triazophos were added to water samples, vegetables and food samples. The triazophos residues were detected by GC-MS. The recoveries of the samples were 65% 125%, coefficient of variation <15%. The sensitivity, specificity, precision, accuracy and validity of the kit were determined. The results showed that the lowest detection limit of the kit was 4.13ng / mL and the linearity range was 3.9-500ng / mL. ℃ kit conditions valid for up to 6 months. Testing triazophos residues with ELISA kit has the characteristics of rapid, simple, sensitive and economical analysis of samples, which can be widely applied to the production of environmental monitoring and safe food.
