目的探讨卵泡抑素(FS)在人肺腺癌细胞中的表达及其作用。方法免疫细胞化学染色检测肺腺癌细胞FS表达情况,细胞计数试剂盒测定细胞增殖,流式细胞术检测细胞凋亡。结果免疫细胞化学染色显示人肺腺癌细胞表达FS;40、50 ng/mL激活素A组肺腺癌细胞增殖〔(吸光度值A分别为(0.49±0.07)及(0.42±0.08))明显低于对照组(0.62±0.09);如果预先用抗FS单克隆抗体中和内源FS后,10 ng/mL激活素A也能抑制肺腺癌细胞增殖(0.48±0.06),由其诱导的肺腺癌细胞凋亡率(24.8±6.7)%明显高于单独激活素A组细胞凋亡率(7.3±2.4)%。结论肺腺癌细胞表达的FS有利于肺腺癌细胞生存,阻断FS分泌或其生物学作用对治疗肺腺癌可能具有重要意义。
Objective To investigate the expression of follistatin (FS) in human lung adenocarcinoma cells and its role. Methods Immunocytochemical staining was used to detect the expression of FS in lung adenocarcinoma cells. Cell proliferation was measured by cell counting kit and apoptosis was detected by flow cytometry. Results The results of immunocytochemical staining showed that the expression of FS in human lung adenocarcinoma cells was significantly lower than that in the control group (the values of A were (0.49 ± 0.07) and (0.42 ± 0.08), respectively) in 40 and 50 ng / mL activin A groups (0.62 ± 0.09); 10 ng / mL activin A also inhibited the proliferation of lung adenocarcinoma cells (0.48 ± 0.06) after the endogenous FS was neutralized with anti-FS monoclonal antibody in advance, and the lungs induced by it Apoptosis rate of adenocarcinoma cells (24.8 ± 6.7)% was significantly higher than that of activin A alone group (7.3 ± 2.4)%. Conclusion The expression of FS in lung adenocarcinoma cells is beneficial to the survival of lung adenocarcinoma cells. It may play an important role in the treatment of lung adenocarcinoma by blocking the secretion of FS or its biological function.
