目的 :观察腺病毒重组肿瘤坏死因子 (TNF)基因转染对大鼠乳腺癌细胞 Walker2 5 6体内致瘤性的影响及瘤体内注射 TNF重组腺病毒对 Walker2 5 6荷瘤大鼠的治疗作用。 方法 :Walker2 5 6乳癌细胞感染人 TNF重组腺病毒后 ,观察其体外增殖能力、皮下致瘤性的变化及进行肿瘤局部病理学检查 ;瘤体局部注射 TNF重组腺病毒 (Ad.h TNF) ,观察荷瘤大鼠的肿瘤生长情况及生存期。结果 :Walker2 5 6感染人 TNF重组腺病毒后 ,培养上清中 2 4h TNF的分泌量达 5 .2 ng/10 6细胞 ,细胞形态学无明显变化 ,体外增殖能力和皮下致瘤性显著下降 ,瘤体周围有大量淋巴细胞及单核细胞浸润 ;肿瘤局部注射 Ad.h TNF能显著抑制 Walker2 5 6的生长 ,并延长荷瘤大鼠的生存期。结论 :重组腺病毒能有效介导 TNF基因转染 Walker2 5 6细胞 ,瘤体内局部注射 Ad.h TNF对 Walker2 5 6荷瘤大鼠具有明显的治疗作用。
OBJECTIVE: To observe the effect of adenovirus recombinant tumor necrosis factor (TNF) gene transfection on in vivo tumorigenesis of rat breast cancer cell line Walker2 5 6 and the therapeutic effect of TNF recombinant adenovirus injected in vivo on Walker 256 tumor-bearing rats. Methods: Walker2 5 6 breast cancer cells were infected with recombinant adenovirus of human TNF and their proliferative capacity and subcutaneous tumorigenicity in vitro were observed. Local tumor pathological examination was performed. TNF-α recombinant adenovirus (Ad.h TNF) The tumor growth and survival of tumor-bearing rats were observed. Results: After infected with human TNF recombinant adenovirus, the secretion of TNF in culture supernatant reached 5.2 ng / 106 cells at 24 h, the cell morphology did not change significantly, and the proliferation ability and subcutaneous tumorigenicity were significantly decreased , A large number of lymphocytes and mononuclear infiltration around the tumor body; local injection of Ad.h TNF can significantly inhibit the growth of Walker2 5 6, and prolong the survival of tumor-bearing rats. CONCLUSION: Recombinant adenovirus can effectively transduce TNF gene into Walker2 5 6 cells. Local injection of Ad.h TNF in tumor tissue has significant therapeutic effect on Walker 256 tumor-bearing rats.
