应用细胞培养、核酸分子原位杂交、免疫细胞化学、图像分析技术研究低氧对肺血管周细胞转化生长因子TGF-β1mRNA及蛋白表达的影响,探讨低氧条件下肺血管周细胞增殖、向平滑肌样细胞分化的机制。结果发现低氧(H)组周细胞内TGF-β1mRNA及蛋白表达量分别是常氧(N)组的1.35倍和1.23倍,差异均有显著意义(均为P<0.01)。研究表明,低氧可促进脉血管周细胞TGF-β1mRNA及蛋白表达增高。TGF-β1表达增高可能是低氧诱导肺血管周细胞增殖、向平滑肌样细胞分化的机制之一。
The effects of hypoxia on the mRNA and protein expression of transforming growth factor-β1 (TGF-β1) in pulmonary vascular cells were studied by cell culture, nucleic acid in situ hybridization, immunocytochemistry and image analysis. The effects of hypoxia on the proliferation of pulmonary pericytes, Like cell differentiation mechanism. The results showed that the expression of TGF-β1 mRNA and protein in pericytes of hypoxic (H) group was 1.35 and 1.23 times higher than that of normoxia group (P <0.01 respectively) ). Studies have shown that hypoxia can promote the expression of TGF-β1 mRNA and protein in vascular pericytes. Increased expression of TGF-β1 may be hypoxia-induced proliferation of pulmonary vascular cells, to smooth muscle-like cell differentiation mechanism.
