目的 探讨端粒酶在小儿急性白血病中的表达及作为预测病情、判断预后标志物的可能性。方法 采用端粒重复序列扩增 微孔杂交法 (TRAP Kyb) ,检测 16例急性白血病患儿骨髓和外周血单个核细胞的端粒酶活性。对照组为10例非白血病血液病患儿的骨髓及 5例正常健康小儿的外周血单个核细胞。结果 16例急性白血病患儿表达阳性率83.1% ) ;10例非白血病血液病患儿有 2例呈低度阳性表达 ,5例正常健康小儿外周血单个核细胞中未见阳性表达。结论 端粒酶活性在急性白血病患儿骨髓和外周血单个核细胞中明显升高 ,表明端粒酶的活性与急性白血病的发生可能有着密切关系 ,检测其活性可能成为一种评估急性白血病病情及预后的有用指标
Objective To investigate the expression of telomerase in children with acute leukemia and to evaluate the prognosis as a prognostic marker. Methods Telomerase activity in bone marrow and peripheral blood mononuclear cells of 16 children with acute leukemia was detected by telomeric repeat amplification microplate hybridization (TRAP Kyb). The control group consisted of 10 patients with peripheral blood mononuclear cells from the bone marrow of children with non-leukemia hematological disease and 5 normal healthy children. Results The positive rate was 83.1% in 16 children with acute leukemia. Two of 10 children with non-leukemia hematologic disease showed low positive expression, while 5 normal healthy children showed no positive expression in peripheral blood mononuclear cells. Conclusion Telomerase activity was significantly increased in the bone marrow and peripheral blood mononuclear cells of children with acute leukemia, indicating that telomerase activity may be closely related to the occurrence of acute leukemia.
