目的研究冬凌草甲素抑制人胃癌SGC-7901细胞生长作用及G2/M期阻滞机制。方法MTT法检测冬凌草甲素对SGC-7901细胞生长的抑制作用;流式细胞术检测冬凌草甲素对SGC-7901细胞周期的影响;West-ern blotting法检测冬凌草甲素对Cdk1、Cyclin B1两种蛋白表达的影响。结果冬凌草甲素作用于SGC-7901细胞的IC50为15.64μmol/L;冬凌草甲素对G2/M期细胞的阻滞作用随着浓度的增加而增强;Cdk1和Cyclin B1蛋白的表达量随着冬凌草甲素浓度的增大显著降低。结论冬凌草甲素通过下调SGC-7901细胞内Cdk1、CyclinB1两种蛋白的表达,阻滞细胞于G2/M期而抑制SGC-7901细胞生长。
Objective To investigate the inhibitory effect of oridonin on the growth of human gastric cancer cell line SGC-7901 and the mechanism of G2 / M arrest. Methods The inhibitory effect of oridonin on the growth of SGC-7901 cells was detected by MTT assay. The effect of oridonin on the cell cycle of SGC-7901 cells was detected by flow cytometry. The activity of oridonin Cdk1, Cyclin B1 expression of two proteins. Results The IC50 of oridonin on SGC-7901 cells was 15.64μmol / L. The inhibitory effect of oridonin on G2 / M phase cells was enhanced with the increase of concentration. The expressions of Cdk1 and Cyclin B1 The amount of Oridonin decreased with the increase of Oridonin. Conclusion Oridonin inhibits the growth of SGC-7901 cells by down-regulating the expression of Cdk1 and CyclinB1 in SGC-7901 cells and blocking the cells in G2 / M phase.
