目的:研究玉郎伞多糖(YLSP)抗鸭乙型肝炎病毒(duck hepatitis B virus,DHBV)的作用。方法:取1日龄广西麻鸭60只,在麻鸭颈静脉注射0.2 mL强阳性DHBV DNA鸭血清,13 d后分别从其颈静脉采血0.5 mL,分离血清,用普通PCR法筛选出DHBV感染强阳性鸭。然后,将50只DHBV DNA阳性麻鸭随机分为5组:YLSP高、中、低剂量组(5,2.5,1.25 g·kg-1);阿昔洛韦阳性对照组(0.1 mg·kg-1)和模型组。所有给药组均与模型组比较,以明确玉郞伞多糖对DHBV DNA的作用,所以未设置正常组。于给药前(T0),给药7 d(T7),14 d(T14)及停药后3 d(P3)采血,用实时荧光定量PCR法检测鸭血清中DHBV DNA的含量。HE染色观察肝损伤程度。结果:与模型组相比,YLSP各剂量组及阳性对照组在用药后血清中DHBV DNA的含量显著降低,停药后,阳性对照组及YLSP中、低剂量组DHBV DNA含量均有反跳现象。HE染色结果显示YLSP能减轻鸭肝组织的病理损伤程度。结论:YLSP对DHBV具有抑制作用同时对肝损伤具有保护作用。
Objective: To study the effect of YLSP on duck hepatitis B virus (DHBV). Methods: Sixty one-day-old Guangxi ducklings were injected with 0.2 mL of DHBV DNA duck serum with strong positive expression in the jugular vein. After 13 d, 0.5 mL of blood was collected from the jugular vein and the serum was separated. DHBV infection was screened by ordinary PCR Strong positive duck. Then 50 DHBV DNA positive ducks were randomly divided into 5 groups: YLSP high, medium and low dose groups (5,2.5,1.25 g · kg-1); acyclovir positive control group (0.1 mg · kg- 1) and model group. All administration groups were compared with the model group to clarify the role of jade umbrella umbrella polysaccharide on DHBV DNA, so no normal group was set. Blood samples were collected before T0, 7d, 14d (T14) and 3 d after stopping (P3). The content of DHBV DNA in duck serum was detected by real-time fluorescence quantitative PCR. HE staining to observe the degree of liver injury. Results: Compared with the model group, the contents of DHBV DNA in the serum of YLSP groups and the positive control group were significantly decreased after treatment, and the DHBV DNA content of positive control group and middle and low dose YLSP groups rebounded after stopping . HE staining results showed that YLSP can alleviate the pathological damage of duck liver tissue. Conclusion: YLSP can inhibit DHBV and protect liver against liver injury.
