目的:探讨肿瘤细胞中zeb1基因的表达量与肿瘤细胞的迁移能力之间的关系.方法:实时定量PCR方法检测正常胃黏膜上皮细胞GES及四种肿瘤细胞BGC823、SGC7901、A549和HeLa细胞中zeb1基因的表达量;Transwell小室检测五种细胞的迁移能力.结果:在五种细胞中,zeb1在HeLa细胞中表达量最高,BGC823及SGC7901次之,在A549及GES中表达量最低;发生迁移的细胞数目在HeLa细胞中最多,BGC823及SGC7901其次,在A549及GES细胞中最少;线性相关分析表明,zeb1基因的表达量与细胞迁移能力呈正相关(r=0.961,P<0.01).结论:zeb1基因可能促进肿瘤细胞的迁移能力.
OBJECTIVE: To investigate the relationship between zeb1 gene expression and tumor cell migration in tumor cells.Methods: Real-time quantitative PCR was used to detect the expression of zeb1 in normal gastric mucosal epithelial cells GES and four tumor cells BGC823, SGC7901, A549 and HeLa Transwell chamber was used to detect the migration ability of five kinds of cells.Results: Among the five kinds of cells, zeb1 was the highest expression in HeLa cells, followed by BGC823 and SGC7901, and lowest in A549 and GES. The number of cells in HeLa cells was the highest, followed by BGC823 and SGC7901, and the least in A549 and GES cells. The linear correlation analysis showed that the expression level of zeb1 was positively correlated with cell migration (r = 0.961, P <0.01) .Conclusion: Genes may promote tumor cell migration.
