目的探讨血管活性肽中介素1-53对油酸致急性肺损伤(ALI)的保护作用及其可能机制。方法将雄性Wistar大鼠24只按随机数字表法分为正常对照组、油酸组和治疗组,每组8只。尾静脉注射油酸(0.2ml/kg)制备大鼠ALI模型,注射前1h及注射后2h各腹腔注射中介素1-53(每次2nmol/kg)。6h后分别行动脉血气分析检查,观察肺组织形态学变化,测定肺湿干质量比值(W/D)、支气管肺泡灌洗液(BALF)中白细胞计数及中性粒细胞分类;测定肺组织髓过氧化物酶活性(MPO)、丙二醛(MDA)和共轭二烯键(C-diene)含量。结果治疗组动脉血氧分压(PaO2)为[(81.7±4.6)mmHg,1mmHg=0.133kPa],肺W/D比值为5.49±0.63,BALF中白细胞计数及中性粒细胞分别为(57.9±7.4)×104/ml、0.718±0.085,与油酸组[(60.8±3.2)mmHg、6.18±0.34、(122.0±16.6)×104/ml、0.878±0.026]比较差异均有统计学意义(q值分别为16.74、3.43、17.23、6.32,P均<0.05、0.01)。结论中介素1-53能明显降低肺组织的脂质过氧化反应,减轻肺组织的损伤程度,对油酸致大鼠ALI具有一定的保护作用。
Objective To investigate the protective effect of interleukin-1-53 on vasoactive acid-induced acute lung injury (ALI) and its possible mechanism. Methods Twenty-four male Wistar rats were randomly divided into normal control group, oleic acid group and treatment group with 8 in each group. Rat ALI model was induced by tail vein injection of oleic acid (0.2ml / kg). Interleukin 1-53 (2nmol / kg) was injected intraperitoneally 1h before injection and 2h after injection. Six hours later, arterial blood gas analysis was performed to observe the morphological changes of the lung tissue. The W / D ratio of the lungs, the white blood cell count in the bronchoalveolar lavage fluid (BALF) and the neutrophil count were determined. Peroxidase activity (MPO), malondialdehyde (MDA) and conjugated diene (C-diene) content. Results The PaO2 was [(81.7 ± 4.6) mmHg, 1 mmHg = 0.133 kPa] and the lung W / D ratio was 5.49 ± 0.63. The white blood cell count and neutrophil count in BALF were (57.9 ± (P0.01), 7.4 × 104 / ml and 0.718 ± 0.085, respectively, which were significantly different from those in oleic group [(60.8 ± 3.2) mmHg, 6.18 ± 0.34, (122.0 ± 16.6) × 104 / ml, 0.878 ± 0.026] Values were 16.74,3.43,17.23,6.32, P <0.05,0.01). Conclusion Interleukin-1-53 can significantly reduce the lipid peroxidation in lung tissue and reduce the degree of lung injury, which has some protective effects on rats induced by oleic acid.
