清热化瘀Ⅱ号方对大鼠脑缺血再灌注损伤IL-6及IL-8表达的影响

目的:研究清热化瘀Ⅱ号方对大鼠脑缺血再灌注损伤缺血侧皮层白细胞介素-6(IL-6)、白细胞介素-8(IL-8)表达的影响。方法:将成年健康SD大鼠按随机数字表分为两大组,每大组再随机分为4个组,空白组、假手术组、模型组、清热化瘀Ⅱ号方组(QRHYⅡ组)。采用线栓法制备大鼠局灶性脑缺血再灌注模型,除空白组外其余3组分别按缺血再灌注2h后3、6、12、24、48h 5个时间点分为5个亚组。取材后,随机选择一组进行HE染色并观察各组脑组织病理形态学改变,另一组应用放射免疫法检测大鼠缺血侧皮层IL-6、IL-8的表达。结果:HE染色结果:空白组、假手术组均未见神经元损伤,模型组及QRHYⅡ组有不同程度的神经元损伤,QRHYⅡ组在各时间点的神经元受损较模型组组明显减轻(P<0.05);放射免疫法结果:与假手术组比较,模型组及QRHYⅡ组IL-6、IL-8在3h开始明显升高,24h达到高峰,48h后仍维持在较高水平,差异有统计学意义(P<0.05);与模型组比较,QRHYⅡ组随着存活时间的延续,IL-6、IL-8的表达在同一时间点明显下降,具有显著性的差异(P<0.01)。结论:清热化瘀Ⅱ号方可能通过下调炎性因子IL-6、IL-8的释放,减轻炎性级联反应,减轻脑缺血再灌注损伤,发挥神经元保护作用。 Objective: To investigate the effects of Qingre Huayu Recipe (Ⅱ) on the expression of interleukin-6 (IL-6) and interleukin-8 (IL-8) in ischemic cortex of rats with cerebral ischemia-reperfusion injury. Methods: Adult healthy SD rats were divided into two groups according to random number table. Each group was randomly divided into four groups: blank group, sham operation group, model group, qingrehuayu Ⅱ group (QRHYⅡ group) . The focal cerebral ischemia-reperfusion model was established by thread occlusion method. The rats in the other three groups were divided into 5 sub-groups at 3, 6, 12, 24 and 48 hours after ischemia / reperfusion group. After the material was taken, one group was randomly selected for HE staining and histopathological changes in each group were observed. The other group was used to detect the expression of IL-6 and IL-8 in ischemic cortex by radioimmunoassay. Results: The results of HE staining showed that there was no neuronal injury in the blank group and the sham operation group. Neuronal damage was induced in the model group and the QRHYⅡ group. The neuronal damage in QRHYⅡ group was significantly lower than that in the model group The levels of IL-6 and IL-8 in model group and QRHYⅡ group were significantly increased at 3h, peaked at 24h and maintained at a high level after 48h, the difference was Statistical significance (P <0.05). Compared with the model group, the expression of IL-6 and IL-8 in QRHYⅡ group decreased significantly at the same time point with the extension of survival time (P <0.01). Conclusion: Qingre Huayu Decoction can reduce the inflammatory cascades, reduce the damage of cerebral ischemia-reperfusion injury and exert neuronal protective effect by down-regulating the release of inflammatory cytokines IL-6 and IL-8.
