去势雄性大鼠肾上腺皮质束状带和网状带的变化及低雄激素对COX-2信号通路的影响

目的观察去势雄性大鼠肾上腺皮质束状带和网状带的形态结构和功能的变化,探讨低雄激素对环氧化酶2(COX-2)信号通路的影响。方法 30只10周龄雄性SD大鼠随机分为对照组(n=8)、去势组(n=11)和替代组(n=11),对照组大鼠接受假去势手术,去势组大鼠接受去势手术,替代组大鼠在接受去势手术后给予肌肉注射睾酮(十一酸睾酮每月50 mg/kg)进行替代治疗。10周后采集各组大鼠血清,放射免疫分析法测定血清睾酮(T)、皮质酮(F)、促肾上腺皮质激素(ACTH)、黄体生成素(LH)和促卵泡激素(FSH)水平;取大鼠肾上腺皮质组织,HE染色后光学显微镜观察肾上腺皮质束状带和网状带形态学变化,透射电子显微镜观察超微结构改变;采用RT-PCR技术分别检测大鼠肾上腺皮质和胸腹主动脉组织中COX-2 mRNA表达。结果与对照组和替代组比较,去势组大鼠血清T水平显著降低(P<0.01),血清F、ACTH、LH和FSH水平显著升高(P<0.05)。去势组大鼠肾上腺皮质束状带和网状带细胞变大,细胞核增大且常染色质增多,线粒体和滑面内质网也较发达,脂滴明显减少;替代组大鼠肾上腺皮质束状带和网状带的细胞形态和超微结构与对照组相似。去势组大鼠肾上腺皮质组织中COX-2 mRNA表达显著低于对照组和替代组(P<0.05),而主动脉血管组织中COX-2 mRNA表达明显高于对照组和替代组(P<0.05)。结论去势雄性大鼠肾上腺皮质束状带和网状带增生且功能增强,肾上腺皮质组织中COX-2 mRNA表达下调;主动脉血管组织中COX-2mRNA表达上调,可能与低雄激素导致的血管病变有关。 Objective To observe the morphological changes and function of adrenal cords and reticular bands in ovariectomized male rats and to explore the effect of low androgen on cyclooxygenase 2 (COX-2) signaling pathway. Methods Thirty male Sprague-Dawley rats, aged 10 weeks, were randomly divided into control group (n = 8), castration group (n = 11) and replacement group (n = 11) The rats in the group undergoing castration were surgically treated. The rats in the alternative group were given intramuscular testosterone (50 mg / kg / day of undecanoate testosterone) for replacement therapy after undergoing castration surgery. The serum of rats in each group was collected 10 weeks later. The levels of serum testosterone, corticosterone, adrenocorticotropic hormone, luteinizing hormone (LH) and follicle stimulating hormone (FSH) were measured by radioimmunoassay. Adrenal cortex tissues were taken from the rats. The morphological changes of adrenal cords and reticular bands were observed under light microscope with HE staining. The ultrastructure of the adrenal cortex was observed by transmission electron microscopy. The adrenal cortex, COX-2 mRNA expression in arterial tissue. Results Compared with the control group and the surrogate group, the serum T level was significantly decreased (P <0.01) and the levels of F, ACTH, LH and FSH were significantly increased in the castration group (P <0.05). Ovariectomized rats adrenal cords and reticular band cells become larger, enlarged nuclei and increased euchromatin, mitochondria and synovial endoplasmic reticulum is also more developed, lipid droplets significantly reduced; instead of rats adrenal cortex bundle The shape and ultrastructure of tapes and reticular tapes were similar to those of the control group. The expression of COX-2 mRNA in adrenocortical tissue of castrated group was significantly lower than that of control group and substitute group (P <0.05), while the expression of COX-2 mRNA in aortic vascular tissue was significantly higher than that of control group and replacement group (P < 0.05). CONCLUSIONS: Cortical and reticular bands of ovariectomized male rats are hyperplastic and have increased function. The expression of COX-2 mRNA in adrenocortical tissues is down-regulated. The expression of COX-2 mRNA in aortic vascular tissues may be up-regulated, Lesions related.
