目的探索FTA卡样本DNA最小检出量及同一FTA卡样本进行多项目检测的可行性。方法制作含有不同浓度DNA FTA卡,使用Identifiler试剂盒进行检验;对同一FTA卡样本先后使用Identifiler、Y-filer试剂盒进行扩增及线粒体高变区HVI区测序,使用ABI3130测序仪检测各扩增产物。结果载有0.5ngDNA的FTA卡扩增产物即可正确分型,对同一血痕样本使用不同试剂盒检验,均可清晰正确判型,线粒体高变区HVI区测序图清晰,且各检验结果均与对照一致。结论载有0.5ngDNA的FTA卡扩增产物可用于DNA分型检测,FTA卡对DNA结合较稳定,可用于多项目检测。
Objective To explore the feasibility of multi-project detection of the minimum detectable amount of DNA in the FTA card sample and the same FTA card sample. Methods DNA FTA cards containing different concentrations of DNA were prepared and verified by using Identifiler kit. The samples of the same FTA card were amplified by using Identifiler, Y-filer kit and HVI region of mitochondrial hypervariable region, and each amplification was detected by ABI3130 sequencer product. The results contain 0.5ngDNA FTA card amplification products can be the correct type, the same bloodstain samples using different kits test, can be clear and correct classification, mitochondrial hypervariable region HVI region sequencing clear, and the test results were Consensus. Conclusion The FTA card containing 0.5ng DNA can be used for DNA typing. The FTA card is more stable to DNA binding and can be used for multi-project detection.
