采用 CTAB方法及其改进方法对枇杷 1 4个品种的基因组 DNA进行了分离、提取和琼脂糖凝胶电泳 ,结果表明 ,采用两种提取法的 1 4个枇杷品种基因组 DNA均有二条清晰泳带 ,其基因组 DNA大小约 2 0 kb;将所分离的基因组 DNA通过紫外分光光度计进行浓度测定 ,结果显示 ,不同枇杷品种 DNA含量存在一定差异 ,浓度介于 2 5.6~ 396.74 μg/ml,其中太城 4号最高 (为 396.74 μg/ml) ,夹脚最低 ( 2 5.2 6μg/m l)。实验还表明 ,改进后的方法 ,提取基因组 DNA纯度高 ,且 A2 60 nm/A2 80 nm的比值均介于 1 .8~ 2 .0。
The genomic DNA of 14 loquat cultivars was isolated and extracted by agarose gel electrophoresis using CTAB method and its improved method. The results showed that there were two clear bands in genomic DNA of 14 loquat cultivars , And its genomic DNA was about 20 kb in size. The isolated genomic DNA was determined by UV spectrophotometer. The results showed that there were some differences in the DNA content of different loquat cultivars at a concentration ranging from 2. 5.6 to 396.74 μg / ml, of which too City 4 was the highest (396.74 μg / ml) with the lowest pinch (2 5.2 6 μg / ml). Experiments also show that the improved method, the extraction of genomic DNA purity, and A260nm / A280nm ratio were between 1.8 ~ 2.0.
