用免疫电镜技术观察了阿片μ受体(MOR)在猫脊髓背角浅层内的超微定位分布。电镜下观察发现MOR样免疫反应产物主要位于小树突和无髓纤维内,偶见有阳性的胞体。MOR阳性树突多与阴性轴突终末形成对称性突触,个别为非对称性突触。背角浅层内分布有一定数量的MOR阳性无髓纤维,但仅观察到少量阳性轴突终末,且其中大部分并不与其周围结构形成突触连接。本研究结果为MOR在脊髓水平参与阿片类物质镇痛过程中存在突触前和突触后两种机制提供了明确的形态学证据。
The ultrastructure of opioid receptor (MOR) in the superficial layer of cat spinal dorsal horn was observed by immunoelectron microscopy. Electron microscopy showed MOR-like immunoreactive products were mainly located in the small dendritic and non-myelinated fibers, occasionally positive cell bodies. MOR-positive dendrites and negative axonal terminals to form symmetrical synapses, some of the asymmetric synapses. A certain number of MOR positive myelinated fibers were distributed in the superficial dorsal horn, but only a few positive axon terminals were observed, and most of them did not form synaptic connections with the surrounding structures. The results of this study provide clear morphological evidence for the existence of both pre-synaptic and post-synaptic mechanisms of MOR during spinal cord-level opioid analgesia.
