目的探讨我国汉族人群p38βMAPK基因多态性对mRNA表达的影响。方法纳入正常人群98名,经测序分析rs2072878位点AA基因型82名,AG基因型16名。分别选取AA与AG基因型组各14名,提取外周血单核细胞(PBMC),提取RNA与蛋白。p38βMAPK mRNA的测定采用实时荧光定量PCR,p38 MAPK总蛋白表达的检测采用SDS-PAGE电泳-蛋白免疫印迹法。结果 p38βMAPK rs2072878的AA基因型人群mRNA表达水平显著低于AG基因型人群[(0.485±1.143)vs(2.250±2.719),P=0.034]。p38MAPK蛋白表达水平AA基因型组与AG基因型组差异无统计学意义[(1.347±0.309)vs(1.007±0.500),P=0.375]。结论我国人群p38βMAPK rs2072878基因态性可能影响其mRNA表达,而对p38MAPK总蛋白的表达没有影响。
Objective To investigate the effect of p38βMAPK gene polymorphism on mRNA expression in Chinese Han population. METHODS: A total of 98 normal subjects were enrolled in this study. A total of 82 AA genotypes and 16 genotypes of rs2072878 were sequenced. Fourteen AA and AG genotype groups were selected respectively, and peripheral blood mononuclear cells (PBMC) were extracted and RNA and protein were extracted. The p38βMAPK mRNA was determined by real-time fluorescence quantitative PCR, and the total protein expression of p38 MAPK was detected by SDS-PAGE electrophoresis-Western blotting. Results The mRNA level of AA genotype of p38βMAPK rs2072878 was significantly lower than that of AG genotype ([0.485 ± 1.143] vs (2.250 ± 2.719), P = 0.034]. There was no significant difference in p38MAPK protein level between AA genotype group and AG genotype group [(1.347 ± 0.309) vs (1.007 ± 0.500), P = 0.375]. CONCLUSION: The gene status of p38βMAPK rs2072878 may affect its mRNA expression in Chinese population, but has no effect on the expression of p38MAPK protein.
