目的通过检测微小miR-340-5p在肺癌患者血清中的表达,研究其与肺癌患者临床病理特征的相关性,并探讨其能否作为肺癌早期诊断的潜在肿瘤标志物的可能性。方法应用逆转录定量聚合酶链反应(RT-q PCR)分析法对55例肺癌患者、21例健康者、32例肺部良性疾病患者血清中的miR-340-5p水平进行检测,采用方差方程的Levene检验及均值方程的t检验进行统计学比较。用ROC曲线对指标的诊断效能进行分析和评价。结果肺癌患者组血清中的miR-340-5p肿瘤标志物水平明显高于健康组和肺部良性疾病患者组,FC值为6.48,差异有统计学意义(F=12.23,P<0.05;t=-4.44,df=83.8,P<0.05)。ROC曲线下面积AUC为0.785,灵敏度为66.3%,特异度为73.7%。结论 miR-340-5p在肿瘤患者中是高表达,在血清中的检测对肺癌的诊断有一定的临床价值,并有较高的灵敏度和特异度,是诊断肺癌较为理想的指标。
Objective To detect the expression of miR-340-5p in the serum of patients with lung cancer, to study its correlation with the clinicopathological features of lung cancer patients, and to explore whether it can be used as a potential tumor marker for early diagnosis of lung cancer. Methods Reverse transcription quantitative polymerase chain reaction (RT-q PCR) was used to detect the level of miR-340-5p in serum from 55 patients with lung cancer, 21 healthy subjects, and 32 patients with benign lung diseases. The variance equation was used. The Levene test and the t-test of the mean equation were compared statistically. The ROC curve was used to analyze and evaluate the diagnostic effectiveness of the indicators. Results The serum levels of miR-340-5p in lung cancer patients were significantly higher than those in healthy and benign lung patients. The FC value was 6.48, and the difference was statistically significant (F=12.23, P<0.05; t= -4.44, df=83.8, P<0.05). The area under the ROC curve has an AUC of 0.785, a sensitivity of 66.3%, and a specificity of 73.7%. Conclusion miR-340-5p is highly expressed in cancer patients. Detection of serum miR-340-5p has certain clinical value in the diagnosis of lung cancer, and has high sensitivity and specificity. It is an ideal indicator for the diagnosis of lung cancer.
