目的建立HPLC法同时测定天山岩黄芪中4种异黄酮的含有量。方法天山岩黄芪70%乙醇提取液的分析采用YMC-C18色谱柱(4.6 mm×250 mm,5μm);以乙腈-0.2%甲酸为流动相,梯度洗脱;体积流量0.8 mL/min;检测波长254 nm;柱温35℃。结果毛蕊异黄酮苷、芒柄花苷、毛蕊异黄酮、芒柄花素分别在1~50、2~70、0.8~50、2~70μg/mL范围内呈良好的线性关系(r=0.999 9),平均加样回收率95.1%~104.1%,RSD 0.5%~2.8%。结论该方法稳定可靠,可用于天山岩黄芪的质量控制。
Objective To establish an HPLC method for simultaneous determination of four isoflavones in Astragalus membranaceus Tianshan. Methods The extract of Astragalus mongholicus 70% ethanol was separated on a YMC-C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile-0.2% formic acid as mobile phase with a gradient of 0.8 mL / min. The detection wavelength 254 nm; column temperature 35 ℃. Results The results showed that there was a good linear relationship (r = 0.999 9) in the ranges of 1 ~ 50,2 ~ 70,0.8 ~ 50 and 2 ~ 70μg / mL for the constituents of calycosin, The average recovery was 95.1% ~ 104.1%, RSD 0.5% ~ 2.8%. Conclusion The method is stable and reliable and can be used for quality control of Astragalus membranaceus in Tianshan Mountains.
