目的 :探讨抗骨质疏松新药XW6 30促进成骨作用的机理。方法 :提取并标记制备TGF_β1和ColⅠ基因的cDNA探针 ,将 3种不同药物作用于大鼠成骨细胞 2 4、72h后 ,提取细胞总RNA ,并与TGF_β1和ColⅠ基因的cDNA探针进行狭缝杂交。结果 :XW6 30具有促进大鼠成骨细胞中TGF_β1和ColⅠmRNA表达的作用 ,在相同浓度 (10 -6mol/L)下 ,其作用优于四环素加哌嗪雌酮和单纯雌酚酮 (P <0 0 5 )。结论 :XW6 30可能通过促进TGF_β1和ColⅠmRNA的表达而促进成骨作用
Objective: To explore the mechanism of anti-osteoporosis XW6 30 in promoting osteogenesis. Methods: cDNA probes of TGF-β1 and ColⅠ gene were prepared and labeled. After three different drugs were applied to rat osteoblasts for 24 and 72 hours, total cellular RNA was extracted and cDNA probes of TGF-β1 and ColⅠ gene were cloned Crossbreed. Results: The effect of XW630 on the expression of TGF-β1 and ColⅠmRNA in rat osteoblasts was better than that of tetracycline plus piperazinone and estrone (P <0 0) at the same concentration of 10 -6 mol / L 0 5). Conclusion: XW630 may promote osteogenesis through promoting the expression of TGF-β1 and ColⅠmRNA
