研究中获得了龟鳖类18个个体长度为660bp的COI基因序列。利用MEGA软件采用P-距离法计算龟鳖类种间及种内遗传距离并构建序列间的UPGMA和邻接系统发生树。结果显示,龟鳖类的种间遗传距离显著大于种内遗传距离。在系统树中,龟鳖类每一物种的个体分别形成各自独立的分支。基于COI基因的DNA条形码在识别龟鳖类物种方面和传统形态学基本一致,而且该基因可以探讨龟鳖类种间的系统发育关系,以COI基因作为DNA条形码,对龟鳖类进行物种鉴定具有一定的可行性。
In this study, COI gene sequences of 660 bp were obtained from 18 individuals of turtles. The MEGA software was used to calculate the intraspecies and intraspecific genetic distance of the turtles using P-distance method and the UPGMA and the adjacent phylogenetic trees were constructed. The results showed that the genetic distance between species of turtles was significantly greater than the intraspecific genetic distance. In the phylogenetic tree, individuals of each species of turtles form their own independent branches. DNA barcodes based on COI gene are basically the same as traditional morphological characters in identifying turtles and species, and the gene can be used to explore the phylogenetic relationship among species of turtles. COI gene is used as DNA barcodes to identify species of turtles. A certain feasibility.
