目的:研究不同温度的顺铂对人卵巢癌细胞株SKOV3的增殖抑制效应及S100A4 mRNA表达的影响。方法:以体外培养的人卵巢浆液性乳头状囊腺癌细胞株SK-OV3为研究对象,按不同的热疗温度,用四甲基偶氮唑蓝(MTT)法测定各组细胞抑制率;流式细胞仪(FCM)分析细胞周期分布;用半定量逆转录聚合酶链反应(RT-PCR)技术检测各组S100A4 mRNA的表达。结果:(1)各组细胞抑制率均较阴性对照组增加(P<0.05),43℃组作用最明显(P<0.01);(2)FCM分析细胞周期显示各组细胞与对照组比较,G0/G1期细胞比例增高,S期细胞比例下降(P<0.05),细胞生长阻滞于G0/G1期;(3)RT-PCR结果显示,不同温度作用后S100A4 mRNA的表达均低于阴性对照组(P<0.01),以43℃组表达最低。结论:不同温度的顺铂热化疗均可抑制卵巢癌细胞株SKOV3增殖及下调S100A4 mRNA的表达,从而抑制其侵袭转移,以43℃时作用最显著。
AIM: To investigate the effects of different concentrations of cisplatin on the proliferation of human ovarian cancer cell line SKOV3 and the expression of S100A4 mRNA. Methods: Human ovarian serous papillary cystadenocarcinoma cell line SK-OV3 was cultured in vitro. The inhibition rate of each group was determined by MTT assay at different hyperthermia temperature. The cell cycle distribution was analyzed by flow cytometry (FCM). The expression of S100A4 mRNA in each group was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results: (1) Compared with the negative control group, the inhibitory rate of each group increased (P <0.05), and the effect was most obvious at 43 ℃ (P <0.01). (2) The cell cycle of FCM showed that compared with the control group, The percentage of cells in G0 / G1 phase increased, the proportion of S phase cells decreased (P <0.05), and the cell growth arrested in G0 / G1 phase. (3) The results of RT-PCR showed that the expression of S100A4 mRNA was lower than the negative The control group (P <0.01), the lowest expression at 43 ℃. CONCLUSION: Chemotherapy with cisplatin at different temperatures can inhibit the proliferation and down-regulate the expression of S100A4 mRNA in ovarian cancer cell line SKOV3 and thus inhibit its invasion and metastasis.
